A group of eminent scientists led by Dr. Jeremy Stone scrambles to a secure laboratory and tries first to isolate the life form while determining why two people from Piedmont - an old alcoholic and a six-month-old baby - survived. The scientists methodically study the alien life form, unaware that it has already mutated and presents a far greater danger in the lab, which is equipped with a nuclear self-destruct device designed to prevent the escape of dangerous biological agents.

A hallmark of the series shows Briggs or Phelps receiving his instructions on a recording that then self-destructs, followed by the theme music composed by Lalo Schifrin. The series aired on the CBS network from September 1966 to March 1973, then returned to television for two seasons on ABC, from 1988 to 1990, retaining only Graves in the cast. Frank Sutton plays Gomer's high-octane, short-fused Gunnery Sergeant Vince Carter, and Ronnie Schell plays Gomer's friend Gilbert "Duke" Slater. Allan Melvin played in the recurring role of Gunnery Sergeant Carter's rival, Sergeant Charley Hacker. The series never discussed nor addressed the then-current Vietnam War, instead focusing on the relationship between Gomer and Sergeant Carter. The show retained high ratings throughout its run.

The series was inspired by the 1950 film Wagon Master directed by John Ford and starring Ben Johnson, Harry Carey Jr. and Ward Bond, and harkens back to the early widescreen wagon train epic The Big Trail starring John Wayne and featuring Bond in his first major screen appearance playing a supporting role. Horton's buckskin outfit as the scout in the first season of the television series resembles Wayne's, who also played the wagon train's scout in the earlier film.
